چکیده

As leukocytes can penetrate into deep regions of a tumor mass, leukocyte-mimetic liposomes (LM-Lipo) containing leukocyte membrane proteins are also expected to tumors by exerting properties those proteins. The aim the present study was examine whether LM-Lipo, which were recently demonstrated actively pass through inflamed endothelial layers, spheroids, and investigate potential LM-Lipo for use as an anticancer drug carrier. We prepared via intermembrane protein transfer from human leukemia cells; onto determined Western blotting. significantly high association with lung cancer A549 cells compared plain liposomes, contributed effective anti-proliferative action encapsulated doxorubicin hydrochloride (DOX). Confocal microscopic images showed that but not could efficiently spheroids. Moreover, DOX-encapsulated suppressed spheroid growth. Thus, transferred retained their unique function, allowed efficient penetration similar leukocytes. In conclusion, these results suggest be useful tumor-penetrating delivery system treatment.

The Way that PEGyl-DSPC Liposomal Doxorubicin Particles Penetrate into Solid Tumor Tissue

BACKGROUND For enhancement of drug effectiveness and reduction of drug toxicity, liposomal drugs have been studied in laboratories and clinics for decades. Although the results obtained from in vitro are encouraging, but the results from in vivo tests were not satisfactory. The main reasons for this situation were that we do not have enough information about the way how liposomal particles pene...

Incorporating Cellular Metabolism into Growth Models of Multicellular Tumor Spheroids

Many chemotherapeutic strategies have failed due to pronounced diffusion limitations in tumors induced by poor vasculaturization. Diffusion limitations, present in threedimensional tissue, produce nutrient deficient regions that eventually become necrotic in tumors in vivo. Multicellular spheroids are better models of in vivo tumors than monolayer cultures because they mimic the heterogeneous m...

Loading of doxorubicin into liposomes by forming Mn2+-drug complexes.

A new procedure for loading doxorubicin into large unilamellar vesicles (LUVs) is characterized. It is shown that doxorubicin can be loaded into LUVs composed of sphingomyelin/cholesterol (55:45 mole/mole) in response to a transmembrane MnSO4 gradient in the absence of a transmembrane pH gradient. Complex formation between doxorubicin and Mn2+ is found to be a driving force for doxorubicin upta...

Doxorubicin encapsulated in sterically stabilized liposomes is superior to free drug or drug-containing conventional liposomes at suppressing growth and metastases of human lung tumor xenografts.

Liposomes containing polyethylene glycol-derivatized phospholipids are able to evade the reticuloendothelial system and thereby remain in circulation for prolonged periods. We report here that doxorubicin encapsulated in these sterically stabilized liposomes (S-DOX) suppresses the growth of established human lung tumor xenografts in severe combined immunodeficient (SCID) mice and inhibits the s...
